Active Pharmaceutical Ingredients (Tacrolimus, Sirolimus, Pimecrolimus, Teicoplanin, Polymyxin B, Fidaxomicin, Daptomycin, Dalbavancin, Amphotericin B, Caspofungin, Micafungin, Anidulafungin) A Business Opportunity for Startups

PROJECT DESCRIPTION

A transplanted kidney, an ICU bed with a resistant bacterial infection, a patient with invasive fungal disease: all of these are reliant on a small number of complex molecules. The twelve fermentation-derived APIs listed above are at the very heart of that group and Indian manufacturers have only just started to come into their own.

Why Startups and New Entrepreneurs Should Choose This Industry

Most of the new blood in pharma will be heading straight into the commodity space and fighting on the penny. This basket is a different game. Each product is made by growing a specialist microorganism so the know how is your moat. You end up with far fewer producers, long term buyer relationships and very strong realisations per kg.

The 12 APIs all share one technology platform. You've got a fermentation block, a purification suite and a QC lab that can run those molecules in campaigns, so you're able to spread your fixed costs and add a new molecule without having to rebuild the plant. Demand is very steady too. Transplant patients are on tacrolimus or sirolimus for life and hospitals will buy their antibiotics and antifungals on clinical need rather than on season.

Market Size, Growth and Export Potential

Industry research puts India's API market at circa $4.4bn in 2025 and up to $6.7bn by 2030 (CAGR of just under 8.8%). India is one of the top three API producing nations in the world by volume and a 2025 industry roadmap suggests Indian manufacturers could be in a position to take 20-30% of the API business that global buyers are currently looking to de-risk from China.

The demand for the end products is the same story. The global tacrolimus market was at $7.3bn in 2025 and is projected to grow at around 5.6% per year through to 2035 with North America at about 40%. The wider immunosuppressant market is estimated at around $50bn in 2025 (figures vary by report). The echinocandin antifungals (the family that includes caspofungin, micafungin and anidulafungin) were at around $1.4bn in 2025 with caspofungin accounting for about 45% of that. The peptide antibiotics (which cover daptomycin and dalbavancin) were at around $5.8bn and are expected to be up to $9bn by 2030.

The export opening is just as positive. Indian API manufacturers are already exporting around 50% of their output to the developed world (USA, Europe, and Japan) and the US generic dalbavancin approvals from late 2025 will extend the number of formulators who require a qualified API source. A single plant can become a global supplier with a USFDA DMF, an EDQM CEP or a Japanese master file.

How These APIs Are Made

Seven of the twelve come from fermentation per se: tacrolimus, sirolimus, teicoplanin, polymyxin B, fidaxomicin, daptomycin and amphotericin B. The other five (pimecrolimus, dalbavancin, caspofungin, micafungin and anidulafungin) start as a fermentation product and are then completed with a controlled chemical step.

Step 1: Strain and Seed Culture: A high yielding strain (e.g. Streptomyces for tacrolimus and daptomycin) is reactivated from a frozen vial and then scaled up through flasks and seed fermenters.

Step 2: Production Fermentation:  The culture goes into large stirred fermenters with a specially tuned nutrient medium. It's fermented for several days with temperature, pH, aeration and feed all being controlled.

Step 3: Harvest and Recovery:  The liquid is separated from the cells by filtration or centrifugation and the product is then captured using solvents or adsorbent resins.

Step 4: Chemical Conversion: For the semi-synthetic 5's the fermentation product is subjected to a reaction block e.g. pneumocandin to caspofungin or echinocandin B to anidulafungin.

Step 5: Purification: Prep chromatography is used to remove the related impurities until the required purity is achieved (i.e. meets pharmacopoeial and customer specs).

Step 6: Crystallisation, Drying and Finishing: The purified product is crystallised or precipitated, then vacuum dried or freeze dried, milled, blended and packed in double polybags and drums under controlled clean room conditions. Injectable grade APIs will have tighter endotoxin and particulate control.

Step 7: Quality Control: Each batch is HPLC tested, GC for residual solvent, moisture, microbiological and endotoxin tested and stability studies are running in parallel.

Main Machinery and Equipment

A compact multi-product unit typically needs the following:

  • Seed and production fermenters (stainless steel, stirred, with in-place sterilisation)
  • Media preparation tanks, boiler and steam sterilisation systems
  • Air compressors, sterile air filters and chilled-water utilities
  • Centrifuges, filter presses and ultrafiltration units for broth clarification
  • Adsorption resin columns and solvent extraction vessels
  • Glass-lined and stainless steel reactors for semi-synthetic steps
  • Preparative HPLC and chromatography skids
  • Evaporators, crystallisers and nutsche filter-dryers
  • Vacuum tray dryers or freeze dryers
  • Milling, sifting and blending equipment
  • QC laboratory instruments: HPLC, GC, UV spectrophotometer, KF titrator and microbiology set-up
  • Effluent treatment plant and solvent recovery unit

Government Schemes and Incentives

India is very much in favour of bulk drug manufacturing and there are a number of routes which would be suitable for this project.

  • Scheme for promotion of Bulk Drug Parks: shared effluent treatment, steam and testing facilities in the notified parks in Gujarat, Himachal Pradesh and Andhra Pradesh thereby reducing the set up cost.
  • PLI Scheme for bulk drugs: incentives for incremental sales of fermentation based KSMs, intermediates and APIs. Product eligibility is notified by the Department of Pharmaceuticals so you do need to confirm your molecule prior to applying.
  • Strengthening of Pharmaceutical Industry scheme: technology upgradation support for MSME units moving to the revised Schedule M and WHO-GMP standards.
  • CGTMSE: collateral free credit cover for eligible MSMEs with term loans available through SIDBI and banks.
  • Startup India: DPIIT recognition, tax benefits and access to funding for innovation led ventures.
  • State industrial policies: capital subsidy, SGST reimbursement, stamp duty and power tariff concessions in the states of Gujarat, Andhra Pradesh, Telangana and Himachal Pradesh.

Major Players

In India Concord Biotech, Biocon and Hetero are the big names in the fermentation and complex APIs space. Concord Biotech has a volume share in excess of 20% in a number of its fermentation APIs and supplies over 70 countries. Overseas the key suppliers are Zhejiang Hisun and Hainan Poly Pharm in China and Xellia in Europe. The demand for the formulations from the originators (Astellas, Merck and Pfizer) has been the driver for this.

The basket rewards the entrepreneur who gets their house in order. Start with 2 or 3 molecules, build a solid cGMP base, file your dossiers early and the rest of the portfolio will follow.
